DMXL2 also known as rabconnectin-3α is a large protein with a mass of approximately 350 kDa. It contains multiple WD40 repeats which suggests a role in protein-protein interactions. DMXL2 is expressed in various tissues with higher concentrations in the brain pituitary gland and reproductive organs. It functions mechanically in intracellular transport and signal transduction influencing neurotransmitter release and hormone secretion.
DMXL2 plays a role in cellular homeostasis and communication. It is part of a protein complex associating with rab3 GTPase-activating proteins to regulate synaptic vesicle exocytosis. This protein contributes to the balance of Ca2+ dynamics in neuronal cells therefore influencing neurotransmitter release processes. Because it interacts directly with components involved in signaling DMXL2 impacts cellular responses to external stimuli.
DMXL2 participates in the Notch signaling pathway and mTOR pathway. This protein interacts with key molecules such as rab3 a small GTPase essential for vesicle trafficking. The contribution of DMXL2 to these pathways enables it to orchestrate critical steps in cell growth proliferation and survival. By engaging in these pathways DMXL2 influences cellular metabolism and synaptic plasticity.
DMXL2 shows a connection to polycystic ovary syndrome (PCOS) and neurodegenerative disorders like Alzheimer's disease. Altered DMXL2 expression or function impacts reproductive endocrinology pathways related to PCOS where it can interact with proteins like FSH receptor. In Alzheimer's disease DMXL2's role in synaptic function ties it to amyloid-beta peptide processes an important aspect of the disease's progression. All lanes: Western blot - Anti-DMXL2 antibody (ab240690) at 1/1000 dilution
Lane 1: HeLa (human epithelial cell line from cervix adenocarcinoma) whole cell lysate at 50 µg
Lane 2: HEK-293T (human epithelial cell line from embryonic kidney transformed with large T antigen) whole cell lysate at 50 µg
Lane 3: Jurkat (human T cell leukemia cell line from peripheral blood) whole cell lysate at 50 µg
Developed using the ECL technique.

DMXL2 immunoprecipitated from Jurkat (human T cell leukemia cell line from peripheral blood) whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) prepared using NETN lysis buffer. Western blot was performed from the immunoprecipitate using ab240690 at 1/100 dilution.
Lane 1: ab240690 used for IP at 20 μl per reaction in Jurkat whole cell lysate.
Lane 2: Control IgG iP in Jurkat whole cell lysate.
Detection: Chemiluminescence with an exposure time of 30 seconds.
