Supplementary information

DNA Polymerase iota also known as POLI is a specialized DNA polymerase involved in DNA repair processes. It is characterized by its ability to catalyze translesion synthesis permitting DNA replication across lesions that stall the replication fork. Mechanically POLI incorporates nucleotides opposite certain types of DNA damage albeit with higher error rates compared to other polymerases. It is encoded by the POLI gene which in humans is located on chromosome 18. The mass of the protein approximates 83kDa. DNA Polymerase iota is expressed primarily in the testis and is found in other tissues but in lower amounts. Its localization and activity underline its unique roles in maintaining genome stability under conditions that challenge the DNA replication machinery.
Biological function summary

DNA Polymerase iota participates in the repair of DNA through its function in translesion synthesis allowing cells to replicate their DNA in the presence of specific types of damage. It operates independently and it does not form a stable complex like some other polymerases such as those in the DNA polymerase zeta pathway. Its incorporation of nucleotides can lead to mutations due to its lower fidelity which can be both beneficial in tolerating damage and risky due to potential mutagenesis. This trade-off is critical for its biological role particularly during spermatogenesis where rapid proliferation occurs.

Pathways

DNA Polymerase iota is an important player in the DNA damage tolerance pathway specifically in the translesion synthesis process. It functions in coordination with other members of the Y-family of DNA polymerases including DNA Polymerase eta (POLH) and kappa (POLK) which contribute to the bypassing of DNA lesions. These interactions allow cells to survive DNA damage that is beyond immediate repair and continue replication though it may introduce mutations. POLI's activities are vital in the damage bypass pathway that complements the nucleotide excision repair process.

DNA Polymerase iota links to an increased risk of certain cancers due to its error-prone DNA synthesis. Its activity may lead to mutations that drive carcinogenesis when coupled with other genetic mutations or environmental factors. It has significant relevance in conditions related to genomic instability such as skin cancer in relation to ultraviolet-induced DNA damage. Moreover POLI’s role correlates with XPC a DNA damage recognition protein in disorders such as Xeroderma pigmentosum. Dysfunction in these pathways that involve POLI highlights its engagement in maintaining genomic integrity and the balance between DNA repair accuracy and tolerance of lesion persistence.

Target data

Error-prone DNA polymerase specifically involved in DNA repair (PubMed : 11013228, PubMed : 11387224). Plays an important role in translesion synthesis, where the normal high-fidelity DNA polymerases cannot proceed and DNA synthesis stalls (PubMed : 11013228, PubMed : 11387224, PubMed : 14630940, PubMed : 15199127). Favors Hoogsteen base-pairing in the active site (PubMed : 15254543). Inserts the correct base with high-fidelity opposite an adenosine template (PubMed : 15254543). Exhibits low fidelity and efficiency opposite a thymidine template, where it will preferentially insert guanosine (PubMed : 11013228). May play a role in hypermutation of immunoglobulin genes (PubMed : 12410315). Forms a Schiff base with 5'-deoxyribose phosphate at abasic sites, but may not have lyase activity (PubMed : 11251121, PubMed : 14630940).
