Immunoprecipitation - Anti-DOHH antibody [EPR16730] - BSA and Azide free (AB240975)
DOHH was immunoprecipitated from SH-SY5Y (Human neuroblastoma from bone marrow cells) whole cell extract with ab197587 at 1/50 dilution. Western blot was performed from the immunoprecipitate using ab197587 at 1/1000 dilution. Anti-Rabbit IgG (HRP), specific to the non-reduced form of IgG, was used as secondary antibody at 1/1500 dilution.
Lane 1 : SH-SY5Y whole cell extract 10 μg (Input). Lane 2 : ab197587 IP in SH-SY5Y whole cell extract. Lane 3 : Rabbit monoclonal IgG (ab172730) instead of ab197587 in SH-SY5Y whole cell extract
Blocking and dilution buffer and concentration : 5% NFDM/TBST.
This data was developed using the same antibody clone in a different buffer formulation containing PBS, BSA, glycerol, and sodium azide (ab197587)

Biological function summary
Proteins such as DOHH play important roles in cellular growth and proliferation. DOHH functions by converting inactive eIF5A precursors into their active form which can influence mRNA translation. This enzyme does not form part of a larger complex with other proteins but its activity is essential for the hypusine modification necessary to activate eIF5A directly influencing protein synthesis.
Pathways
DOHH operates in the hypusine biosynthesis pathway which is important for cell proliferation and differentiation. DOHH acts after the enzyme deoxyhypusine synthase (DHS) introduces a deoxyhypusine residue into eIF5A illustrating the close relationship between these two proteins in this cascade. The activation of eIF5A through hypusination impacts diverse pathways that regulate cell cycle progression and apoptosis.
