DOHH was immunoprecipitated from SH-SY5Y (Human neuroblastoma from bone marrow cells) whole cell extract with ab197587 at 1/50 dilution. Western blot was performed from the immunoprecipitate using ab197587 at 1/1000 dilution. Anti-Rabbit IgG (HRP), specific to the non-reduced form of IgG, was used as secondary antibody at 1/1500 dilution.

The deoxyhypusine hydroxylase (DOHH) enzyme also known as hDOHH catalyzes the final step in the post-translational synthesis of the unusual amino acid hypusine. DOHH modifies deoxyhypusine into hypusine by hydroxylation a reaction critical in activating the eukaryotic initiation factor 5A (eIF5A). The human DOHH protein has a mass of approximately 36 kDa and is ubiquitously expressed in various tissues reflecting its general significance in protein processing.
Biological function summary

Proteins such as DOHH play important roles in cellular growth and proliferation. DOHH functions by converting inactive eIF5A precursors into their active form which can influence mRNA translation. This enzyme does not form part of a larger complex with other proteins but its activity is essential for the hypusine modification necessary to activate eIF5A directly influencing protein synthesis.

Pathways

DOHH operates in the hypusine biosynthesis pathway which is important for cell proliferation and differentiation. DOHH acts after the enzyme deoxyhypusine synthase (DHS) introduces a deoxyhypusine residue into eIF5A illustrating the close relationship between these two proteins in this cascade. The activation of eIF5A through hypusination impacts diverse pathways that regulate cell cycle progression and apoptosis.

DOHH activity has implications in cancer and neurodegenerative diseases. Altered expression or malfunction of DOHH can influence eIF5A activity affecting cell growth and survival and therefore contributing to oncogenesis. In neurodegenerative conditions abnormal hypusine synthesis may disrupt neuronal integrity and function. Catalyzes the hydroxylation of the N(6)-(4-aminobutyl)-L-lysine intermediate produced by deoxyhypusine synthase/DHPS on a critical lysine of the eukaryotic translation initiation factor 5A/eIF-5A. This is the second step of the post-translational modification of that lysine into an unusual amino acid residue named hypusine (PubMed : 16371467, PubMed : 16533814, PubMed : 19706422). Hypusination is unique to mature eIF-5A factor and is essential for its function (By similarity).
