Membranous and cytoplasmic staining on human germ cells but not Leydig cells (arrow) or Sertoli cells (arrowhead) (PMID : 21724266) is observed. Counter stained with hematoxylin.

Secondary antibody only control : Used PBS instead of primary antibody, secondary antibody is Goat Anti-Rabbit IgG H&L (HRP) ready to use.

Heat mediated antigen retrieval using ab93684 (Tris/EDTA buffer, pH 9.0).

DPEP3 was immunoprecipitated from 0.35 mg of human testis lysate with ab235451 at 1/30 dilution. Western blot was performed from the immunoprecipitate using ab235451 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366), was used for detection at 1/5000 dilution.

Lane 1 : Human testis lysate 10 μg (Input).
Lane 2 : ab235451 IP in human testis lysate.
Lane 3 : Rabbit monoclonal IgG (ab172730) instead of ab235451 in human testis lysate.

Blocking and dilution buffer and concentration : 5% NFDM/TBST.

Dipeptidase 3 (DPEP3) also known as MBD-2 or membrane-bound dipeptidase 3 is an enzyme that performs important roles in hydrolyzing dipeptides. DPEP3 has a molecular weight of approximately 52 kDa. It belongs to the dipeptidase family and is often found in the membranes of the kidney and testis where it contributes to peptide metabolism. As a metalloprotease DPEP3 requires a metal ion as a cofactor for its catalytic activity.
Biological function summary

DPEP3 contributes to diverse physiological processes in the body. This enzyme often exists as a part of a multi-protein complex facilitating the breakdown of dietary peptides and the renal processing of glutathione and leukotriene D4. Through its enzymatic action DPEP3 helps in managing extracellular matrix homeostasis and detoxifying a wide range of substances that circulate in the body.

Pathways

DPEP3 influences several metabolic pathways by engaging in the breakdown of dipeptides. Of particular note DPEP3 functions within the leukotriene metabolism and glutathione metabolism pathways. In leukotriene metabolism enzymes such as leukotriene A4 hydrolase collaborate with DPEP3 for the conversion of leukotrienes into biologically active lipid mediators. In the glutathione metabolism pathway DPEP3's activity is augmented by its interaction with glutathione S-transferases.

DPEP3 shows relevance to conditions like renal dysfunctions and allergic responses. The enzyme's role in metabolizing leukotrienes links it to asthma and other allergic conditions where dysregulated leukotriene pathways play a part. Furthermore changes in DPEP3 expression might be observed alongside alterations in the activity of glutathione S-transferases which could impact renal health and lead to disease.

Target data

Lacks dipeptidase activity and is unable to hydrolyze cystinyl-bis-glycine, leukotriene D4 and the beta-lactam antibiotic imipenem (PubMed : 32325220). The absence of activity may be due to the inability of asparagine (instead of aspartate found in DPEP1/2) at position 359 to function as the acid/base catalyst and activate the nucleophilic water/hydroxide (PubMed : 32325220). A tyrosine (instead of histidine) at position 269 reduces affinity for the beta zinc and may cause substrate steric hindrance (PubMed : 32325220).
