Supplementary information

The DRAM (Damage-Regulated Autophagy Modulator) protein also known by its alternate name "drum" plays a significant role in the regulation of autophagy. DRAM serves as an effector molecule that interacts with specific molecular pathways to control cell self-digestion processes. Its molecular weight falls within the range typical for small regulatory proteins. Expression of DRAM mainly occurs in various tissues with a higher presence observed in human epithelial tissues.
Biological function summary

DRAM facilitates the autophagy process which is critical for cellular homeostasis and balancing the degradation and recycling of cellular components. It functions closely with other autophagy-related proteins forming part of a complex that includes well-studied regulators like ULK1 and LAMP2. The modulation of autophagic activity by DRAM has implications for both normal cellular processes and the response to cellular stresses.

Pathways

The product of the DRAM gene integrates into the autophagic mechanisms where it acts to enhance degradation of defective proteins and organelles. It has associations within the p53 signaling pathway through which it mediates apoptosis and autophagy. Proteins such as p53 and BCL-2 are closely related to the action of DRAM influencing cellular fate decisions during stress conditions.

DRAM's role in autophagy links it to cancer and neurodegenerative diseases. Upregulated DRAM expression is frequently observed in tumor cells where it may influence cancer progression by modulating cell survival pathways. Additionally dysregulated autophagy involving DRAM has connections to Alzheimer's disease potentially due to interactions with proteins like APP which undergo autophagic degradation processes that this protein modulates.
