DSPP also known as Dentin Sialophosphoprotein is a protein important for tooth development and dentin mineralization. It has a molecular mass of about 110-130 kDa which can vary due to post-translational modifications and cleavage into its components. This protein mainly expresses in odontoblasts the cells responsible for dentin formation but is also found in other tissues like bone and inner ear. The protein shares structural similarities with other members of the SIBLING family which are known to interact with mineralized tissues.
Biological function summary

DSPP plays an essential role in the formation and maintenance of dentin structure. It first undergoes proteolytic cleavage into two main products: Dentin Sialoprotein (DSP) and Dentin Phosphoprotein (DPP). These components integrate into the extracellular matrix where they contribute to the mineralization process of dentin. DSPP does not act alone but participates in a protein complex involving other matrix molecules that help regulate mineralization.

Pathways

DSPP is a component of key mineralization pathways which ensure proper formation and hardening of dentin. One significant pathway is the Transforming Growth Factor-beta (TGF-beta) signaling pathway which influences dental tissue response during tooth development. Within this pathway DSPP interacts with proteins like Bone Morphogenetic Protein (BMP) to facilitate signaling processes required for odontoblast differentiation and function.

Mutations or dysfunctional expression of DSPP link to dental defects such as Dentinogenesis Imperfecta and dentin dysplasia. These conditions lead to abnormal dentin formation causing teeth to be discolored weak or misformed. Additionally DSPP shows interactions with proteins associated with bone disorders which suggests a role in conditions like osteogenesis imperfecta.
