Function

Acts as one of several non-catalytic accessory components of the cytoplasmic dynein 1 complex that are thought to be involved in linking dynein to cargos and to adapter proteins that regulate dynein function. Cytoplasmic dynein 1 acts as a motor for the intracellular retrograde motility of vesicles and organelles along microtubules. The intermediate chains mediate the binding of dynein to dynactin via its 150 kDa component (p150-glued) DCTN1. May play a role in mediating the interaction of cytoplasmic dynein with membranous organelles and kinetochores.

Activity summary

Dynein axonemal intermediate chain sometimes referred to as IC or axonemal dynein plays an important role in cellular motility. This protein functions mechanically as a part of the dynein motor complex facilitating the sliding of microtubules that drive the movement of cilia and flagella. Dynein axonemal intermediate chain has a molecular mass ranging between 70 to 90 kDa making it an essential structural component of the axoneme in eukaryotic cells. This protein is ubiquitously expressed in cells possessing motile cilia and flagella including those in the respiratory tract and reproductive system.

Biological function summary

Dynein axonemal intermediate chain is a critical part of the axonemal dynein complex where it bridges motor proteins to linkers ensuring the mechanical force required for ciliary and flagellar beating. It contributes to the motility of spermatozoa and the clearance of mucus in the respiratory epithelial cells. These activities rely on its interaction with heavy and light chains within the dynein motor complex forming a multi-subunit assembly that co-operatively performs directed movements.

Pathways

Dynein axonemal intermediate chain integrates into cellular signaling pathways that control ciliary beating pattern an important pathway being the intraflagellar transport (IFT) system. This system coordinates with other proteins such as kinesin to regulate the bidirectional transport of ciliary components. The interplay between dynein complexes and IFT is essential for maintaining ciliary function and structure highlighting its pivotal role in motility-associated pathways.

Associated diseases and disorders

Defects in dynein axonemal intermediate chain can lead to conditions such as Primary Ciliary Dyskinesia (PCD) and Kartagener's syndrome which manifest as chronic respiratory infections and infertility due to impaired ciliary function. This protein's dysfunction affects associated dynein proteins disrupting the normal architecture and function of axonemal complexes which contributes to the symptoms observed in these disorders. Addressing defects in this protein and its interactions holds potential for therapeutic advancements in treating ciliary dyskinesia-related ailments.
