E2F2 also known as E2F transcription factor 2 is a member of the E2F family of transcription factors. It is a protein with a mass of approximately 46 kDa. It plays an important role by regulating the expression of genes involved in cell cycle progression particularly the transition from G1 to S phase. Researchers have identified that E2F2 is expressed in various tissues with high expression noted in actively dividing cells such as those in fetal tissues adult testis and tumor cells.
Biological function summary

E2F2 functions as a transcriptional activator and repressor controlling genes necessary for DNA synthesis and cell cycle progression. It often associates with retinoblastoma protein (RB1) to form complexes that finely regulate gene transcription. E2F2 not only controls cell proliferation but also influences cell differentiation processes by coordinating the balance between distinct cellular states.

Pathways

E2F2 interacts with key players in the cell cycle such as cyclins and cyclin-dependent kinases (CDKs). It is an integral part of the Rb-E2F signaling pathway which governs cell cycle checkpoints and progression. This pathway's regulation heavily influences the activities of other E2F family members such as E2F1 and E2F3 which can either promote or inhibit cell cycle transitions in response to various signals.

E2F2's dysregulation associates with cancer and retinoblastoma. Uncontrolled E2F2 activity often contributes to tumorigenesis by bypassing cell cycle controls resulting in unchecked cell division. In cancer altered interactions between E2F2 and proteins like RB1 can lead to the loss of tumor suppressor functions such as those observed in certain aggressive tumors.
