Function

Component of the endosomal sorting complex required for transport II (ESCRT-II), which is required for multivesicular body (MVB) formation and sorting of endosomal cargo proteins into MVBs, and plays a role in autophagy (PubMed:38423010). The MVB pathway mediates delivery of transmembrane proteins into the lumen of the lysosome for degradation. The ESCRT-II complex is probably involved in the recruitment of the ESCRT-III complex. The ESCRT-II complex may also play a role in transcription regulation by participating in derepression of transcription by RNA polymerase II, possibly via its interaction with ELL. Required for degradation of both endocytosed EGF and EGFR, but not for the EGFR ligand-mediated internalization. It is also required for the degradation of CXCR4. Required for the exosomal release of SDCBP, CD63 and syndecan (PubMed:22660413).

Activity summary

EAP30 also known as VPS22 or SNF8 is a component of the ESCRT-II complex with a mass of approximately 30 kDa. It plays an important role in the endosomal sorting process a mechanism that directs the transport of ubiquitinated transmembrane proteins for lysosomal degradation. EAP30 is expressed in various tissues but shows high expression levels in cells involved in membrane trafficking and processes requiring active protein sorting.

Biological function summary

The EAP30 protein functions as part of the ESCRT-II complex which is essential for the formation of multivesicular bodies (MVBs) and their sorting machinery. EAP30 interacts with other components of the ESCRT machinery such as EAP20 and VPS36 ensuring effective processing of cellular signaling receptors and achieving membrane dynamics. This interaction facilitates the correct trafficking of membrane proteins which is necessary for maintaining cellular homeostasis.

Pathways

EAP30 is deeply integrated into the endosomal-lysosomal pathway which is essential for directing proteins toward lysosomal degradation. The protein operates closely with the ESCRT complex including ESCRT-I and ESCRT-III and is involved in endocytosis and receptor downregulation.
