Supplementary information

EBNA 3A also known as Epstein-Barr Nuclear Antigen 3A is a protein associated with the Epstein-Barr virus (EBV). This protein has a mass of approximately 115 kDa and is expressed in host cells infected with EBV. EBNA 3A functions as a transcriptional regulator by interacting with DNA and modulating gene expression. EBNA 3A is critical for viral latency and is expressed within the nuclei of infected B lymphocytes contributing to the persistence of the virus within host cells.
Biological function summary

EBNA 3A modulates the host cell cycle and contributes to the immortalization of infected B cells. EBNA 3A EBNA 3B and EBNA 3C form a complex that regulates viral and cellular genes. This complex plays roles in transcriptional repression and activation affecting cell growth and transformation. Through these mechanisms EBNA 3A influences immune evasion by the virus as EBV can maintain a state of latency and avoid detection by the host immune system.

Pathways

EBNA 3A participates in the regulation of the cell cycle and immune signaling. EBNA 3A affects pathways such as the Notch signaling pathway and can alter interactions with proteins like RBP-Jκ. These pathways are important for the control of gene expression and cell differentiation in infected cells. By influencing these pathways EBNA 3A helps EBV manipulate host cell behaviors to support viral replication and persistence.

EBNA 3A is linked to conditions like Burkitt's lymphoma and Hodgkin's lymphoma. These cancers are associated with the transformation of B cells resulting from EBV infection and EBNA 3A collaborates with other viral proteins such as EBNA-LP to contribute to the oncogenic processes. The presence and activity of EBNA 3A in these lymphomas suggest that targeting this protein could offer potential therapeutic avenues for managing EBV-related malignancies.
