Activity summary

ECEL1 also known as endothelin-converting enzyme-like 1 or puromycin-sensitive aminopeptidase is a zinc-dependent metalloprotease with a molecular mass of about 85 kDa. This enzyme primarily expresses in the nervous system indicating a unique role in neuronal function. ECEL1 localizes in the endoplasmic reticulum and Golgi apparatus participating in processing neuropeptides although its full range of substrates is not completely defined.

Biological function summary

ECEL1 functions to modulate neuropeptide activity by cleaving specific peptide bonds acting as a critical regulator in the nervous system. It does not appear to form part of a larger protein complex but interacts closely with other enzymes involved in neuropeptide processing. Through its enzymatic activity ECEL1 plays a role in maintaining neurotransmitter balance thereby influencing nervous system communication.

Pathways

ECEL1 participates in neuropeptide and signaling pathways such as the endothelin-signaling pathway. ECEL1's activity impacts the function and expression of proteins like endothelin and other neuropeptides involved in vasoconstriction and neurotransmission. ECEL1 works alongside enzymes like neprilysin cooperating in peptide degradation and activation processes within these pathways.

Associated diseases and disorders

ECEL1 has association with musculoskeletal and neurological conditions including congenital contractural arachnodactyly. Mutations in ECEL1 can lead to disruptions in neuropeptide processing contributing to developmental abnormalities. Moreover studies suggest an involvement of ECEL1 with proteins such as those related to collagen metabolism linking it to connective tissue disorders and highlighting its role in proper muscular and neuronal development.
