Immunofluorescence analysis of HeLa cells labelling ECM29 with ab28666 at 1/250 dilution. Cells were fixed with 4% paraformaldehyde for 10 minutes, and blocked with 1% BSA for 1 hour at room temperature. Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate at 1 : 2000 dilution for 45 minutes at room temperature was used as the secondary antibody (green) (a). F-actin (was stained with Rhodamine Phalloidin (red) (c) and Nuclear DNA was labelled with DAPI (blue) (b). (d) represents the merged image showing cytoplasmic localization. ECM29 also known as proteasome-associated protein ECM29 homolog is a multi-domain protein with a molecular mass of approximately 209 kDa. The protein acts as an adaptor that links molecular chaperones to the proteasome. Highly expressed in tissues with high proteasome activity ECM29 mainly locates in the cytosol. The structure of ECM29 suggests it interacts with the proteasomal regulatory particle altering its conformation and functional state.
Biological function summary

ECM29 impacts protein degradation by stabilizing the interaction between the 19S regulatory particle and the 26S proteasome core complex. It plays a role in ensuring the correct assembly of the proteasome which is essential for the ubiquitin-proteasome system. This system is responsible for the majority of protein turnover in the cell impacting various cell cycle processes. ECM29's association with other proteasome components like Rpn1 and Rpn2 signifies its role in protein homeostasis.
