Immunocytochemistry/ Immunofluorescence - Anti-ECM29 antibody (AB28666)
Immunofluorescence analysis of HeLa cells labelling ECM29 with ab28666 at 1/250 dilution. Cells were fixed with 4% paraformaldehyde for 10 minutes, and blocked with 1% BSA for 1 hour at room temperature. Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate at 1 : 2000 dilution for 45 minutes at room temperature was used as the secondary antibody (green) (a). F-actin (was stained with Rhodamine Phalloidin (red) (c) and Nuclear DNA was labelled with DAPI (blue) (b). (d) represents the merged image showing cytoplasmic localization. (e) shows the no primary antibody control. The images were captured at 60X magnification.

Biological function summary
ECM29 impacts protein degradation by stabilizing the interaction between the 19S regulatory particle and the 26S proteasome core complex. It plays a role in ensuring the correct assembly of the proteasome which is essential for the ubiquitin-proteasome system. This system is responsible for the majority of protein turnover in the cell impacting various cell cycle processes. ECM29's association with other proteasome components like Rpn1 and Rpn2 signifies its role in protein homeostasis.
Pathways
ECM29 integrates into the ubiquitin-proteasome pathway and cell cycle regulation mechanisms. It coordinates with proteins such as ubiquitin and Rpn10 to manage protein degradation. Its involvement in NAD(P)H pathway regulation has been noticed where it potentially influences cellular responses to stress by modulating proteasome activity under different conditions.
