The target eEF1A1+eEF1A2+eEF1AL3 also known by its full name as elongation factor 1-alpha consists of three isoforms involved in protein synthesis. These isoforms are encoded by different genes and have masses around 50 kDa each. eEF1A1 is expressed widely in various tissues particularly in proliferating cells while eEF1A2 expression is more restricted to muscle heart and brain tissues and eEF1AL3 is less studied regarding its tissue distribution. Mechanically these proteins play a role in the translation elongation phase facilitating the entry of aminoacyl-tRNA into the ribosome A-site.
EEF1A proteins are essential components of the translational machinery interacting with other translation factors and ribosomal subunits. As part of the eEF1 complex they aid in ensuring the fidelity of protein translation confirming that tRNAs are correctly paired with mRNA codons. These proteins also exhibit roles beyond translation influencing processes such as cytoskeleton organization suggesting that they might impact cell shape and motility.
EEF1A1+eEF1A2+eEF1AL3 participate mainly in protein synthesis pathways such as the mTOR signaling pathway which regulates cell growth and metabolism. They interact with ribosomal proteins and translation initiation factors that keep protein biosynthesis efficient and responsive to cellular needs. The mTOR pathway is closely connected to other pathways like AKT signaling with proteins like TSC1 and TSC2 facilitating interactions within this network.
EEF1A proteins have links to various conditions including cancer and neurodegenerative diseases. Increased eEF1A expression has been observed in certain cancers like ovarian cancer suggesting a role in cell proliferation and survival. In neurodegeneration eEF1A2 is particularly highlighted in disorders like amyotrophic lateral sclerosis (ALS) where its dysregulation may lead to neuronal cell death. The involvement of these proteins in such diverse diseases reflects their critical roles in maintaining cellular function.

ICC/IF image of ab37969 stained human HeLa cells. The cells were PFA fixed (10 min), permabilised in TBS-T (20 min) and incubated with the antibody (ab37969, 1μg/ml) for 1h at room temperature. 1%BSA / 10% normal serum / 0.3M glycine was used to quench autofluorescence and block non-specific protein-protein interactions. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red). DAPI was used to stain the cell nuclei (blue).
