Flow Cytometry (Intracellular) - Anti-EGR2 antibody [EPR23228-40] (AB245228)
Flow cytometry overlay histogram showing treated mouse splenocytes on the right (with 5 µg/ml anti-mouse CD3 and 2 µg/ml CD28 for 16 hours) and untreated negative mouse splenocytes on the left, both stained with ab245228 (red line). The cells were fixed with Cytofix and then permeabilised with 2N HCl and 0.1M Borate buffer. The cells were then incubated in 1x PBS containing 10% mouse serum and 10 % normal goat serum to block FC receptors and non-specific protein-protein interaction followed by the antibody (ab245228) (1x 106 in 100μl at 0.2μg/ml (1/10,500 dilution)) for 30 mins at 22°C.
The secondary antibody Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) preadsorbed was incubated at 1/2000 dilution for 30 mins at 22°C
Isotype control antibody (black line) was Recombinant Rabbit IgG, monoclonal [EPR25A] - Isotype Control used at the same concentration and conditions as the primary antibody. Unlabelled sample (blue line) was also used as a control.
Acquisition of >5000 events were collected using a 50 mW Blue laser (488nm) and 525/40 bandpass filter.

IHC results showed nuclear staining on the rat skin (PMID : 21514423, 21514423).The section was incubated with ab245228 for 30 mins at room temperature. The immunostaining staining was performed on a Leica Biosystems BOND® RX instrument. Counterstained with Hematoxylin.

The cells were incubated in 1x PBS containing 10μg/ml anti CD16/CD32 and 10% normal goat serum to block FC receptors and non-specific protein-protein interactions, followed by the antibody (ab245228) (1x 106 in 100μl at 5 μg/ml (1/100 dilution)) and CD4-APC multistain for 30min on ice.
The secondary antibody Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) preadsorbed was incubated at 1/4000 for 30min on ice.

Intracellular flow cytometric analysis of 2% paraformaldehyde fixed, True-Nuclear buffer-permeabilized Mouse splenocyte (treated with 5ug/ml anti-mouse CD3 and 2ug/ml anti-mouse CD28 for 16h) cells labelling EGR2 with ab245228 at 1/500 dilution (Right) compared with a Rabbit monoclonal IgG (ab172730) isotype control (Left). Goat anti rabbit IgG (Alexa Fluor® 488, ab150077) at 1/2000 dilution was used as the secondary antibody.
Cells were surface stained with anti-CD4 conjugated to APC-Cy7 . Then fixed with 2% PFA for 10min followed by intracellularly stained with rabbit IgG (Left) or ab245228 (Right).

Biological function summary
EGR2 influences the development and function of the peripheral nervous system contributing to processes like nerve myelination. It acts as a transcriptional regulator controlling the expression of genes that are essential for the maintenance of myelin sheaths around nerves. While EGR2 does not directly form part of a protein complex its role as a transcription factor allows it to interact with many other proteins to exert its regulatory functions.
Pathways
EGR2 participates in cellular pathways such as the MAPK/ERK signaling pathway and the immune response pathway. In the MAPK/ERK pathway EGR2 interacts with proteins like ERK1/2 to regulate cell growth and differentiation. In immune response pathways EGR2 modulates the differentiation of T cells impacting immune homeostasis and function by influencing gene expression pertinent to immune regulation.
