EIF3D also known as eukaryotic translation initiation factor 3 subunit D is part of the EIF3 complex and plays a fundamental role in the initiation phase of protein synthesis. EIF3D has a molecular weight of approximately 66 kDa. It is expressed in various tissues with high levels observed in cells that have a high rate of protein synthesis such as those found in proliferating tissues.
Biological function summary

EIF3D integrates into the large EIF3 complex which consists of multiple subunits and is vital for the proper assembly of the 40S ribosomal subunit. This assembly process is essential for the initiation of translation acting as a scaffold for other initiation factors. EIF3D contributes to the stabilization and binding of mRNAs to the ribosome facilitating efficient translation initiation. The protein is integral for mRNA selection and because of its interaction within the complex it influences cellular proliferation and growth.

Pathways

EIF3D participates actively in translation initiation and mRNA surveillance pathways. These pathways regulate gene expression at the translational level maintaining the fidelity and quality control of protein synthesis. EIF3D works closely with EIF3E another subunit within the EIF3 complex to modulate these processes. Their interaction ensures the correct binding of mRNA and aids in the prevention of errors during protein synthesis.

EIF3D has been implicated in cancer due to its high expression in rapidly proliferating cells and its role in sustaining increased protein synthesis rates that tumors demand. Moreover EIF3D relates to viral infections where pathogens exploit the host's translational machinery for their replication. mRNA cap-binding component of the eukaryotic translation initiation factor 3 (eIF-3) complex, a complex required for several steps in the initiation of protein synthesis of a specialized repertoire of mRNAs (PubMed : 27462815). The eIF-3 complex associates with the 40S ribosome and facilitates the recruitment of eIF-1, eIF-1A, eIF-2 : GTP : methionyl-tRNAi and eIF-5 to form the 43S pre-initiation complex (43S PIC). The eIF-3 complex stimulates mRNA recruitment to the 43S PIC and scanning of the mRNA for AUG recognition. The eIF-3 complex is also required for disassembly and recycling of post-termination ribosomal complexes and subsequently prevents premature joining of the 40S and 60S ribosomal subunits prior to initiation (PubMed : 18599441, PubMed : 25849773). The eIF-3 complex specifically targets and initiates translation of a subset of mRNAs involved in cell proliferation, including cell cycling, differentiation and apoptosis, and uses different modes of RNA stem-loop binding to exert either translational activation or repression (PubMed : 25849773). In the eIF-3 complex, EIF3D specifically recognizes and binds the 7-methylguanosine cap of a subset of mRNAs (PubMed : 27462815)..
