Supplementary information

EIF3H also known as eukaryotic translation initiation factor 3 subunit H is a component of the eIF3 complex which plays a substantial role in translation initiation. The protein possesses a molecular mass of approximately 42 kDa. EIF3H is broadly expressed in various tissues and cells including high expression levels in proliferating cells. The protein is part of the larger eIF3 complex which consists of 13 subunits contributing to the formation of the pre-initiation complex essential for mRNA translation.
Biological function summary

EIF3H facilitates the recruitment of mRNA to the ribosome by stabilizing the interaction between mRNA and the 40S ribosomal subunit. It functions within the eIF3 complex to assist in ribosomal scanning and the start codon selection. As part of this complex EIF3H helps govern the efficiency of translation initiation impacting protein synthesis rates. These activities are important for cell growth and protein expression regulation underlying the complex's importance in cellular processes.

Pathways

EIF3H plays a role in the mTOR signaling pathway and the MAPK/ERK pathway both extensively involved in regulating cell growth and proliferation. Through these pathways EIF3H interacts with proteins such as eIF4G and ribosomal protein S6 which help coordinate the translation machinery's function and cellular metabolic control. These pathways highlight EIF3H's role in translational control affecting overall cellular function.

EIF3H is implicated in cancer particularly in types characterized by unchecked cellular proliferation such as lung and breast cancer. EIF3H's dysregulated expression or activity can influence oncogenic pathways leading to malignancies. Additionally EIF3H interacts with proteins like c-Myc a well-known oncogene through its involvement in cancer pathways. These associations highlight EIF3H as a potential target for therapeutic intervention in cancer treatments.

Target data

Component of the eukaryotic translation initiation factor 3 (eIF-3) complex, which is required for several steps in the initiation of protein synthesis (PubMed : 17581632, PubMed : 25849773, PubMed : 27462815). The eIF-3 complex associates with the 40S ribosome and facilitates the recruitment of eIF-1, eIF-1A, eIF-2 : GTP : methionyl-tRNAi and eIF-5 to form the 43S pre-initiation complex (43S PIC). The eIF-3 complex stimulates mRNA recruitment to the 43S PIC and scanning of the mRNA for AUG recognition. The eIF-3 complex is also required for disassembly and recycling of post-termination ribosomal complexes and subsequently prevents premature joining of the 40S and 60S ribosomal subunits prior to initiation (PubMed : 17581632). The eIF-3 complex specifically targets and initiates translation of a subset of mRNAs involved in cell proliferation, including cell cycling, differentiation and apoptosis, and uses different modes of RNA stem-loop binding to exert either translational activation or repression (PubMed : 25849773).
