Supplementary information

The E74-like factor 4 (Elf4) also known as Myeloid Elf-1 like factor (MEF) is a transcription factor belonging to the ETS family. Elf4 is critical for regulating gene expression involved in immune response and hematopoiesis. With a molecular mass of approximately 67 kDa Elf4 is expressed in various tissues especially in hematopoietic cells such as T cells NK cells and myeloid cells. Its expression in these cells highlights its importance in immune system regulation.
Biological function summary

Elf4 functions as a transcriptional activator influencing cell cycle progression and apoptosis. It operates within a complex that includes co-factors necessary for effective transcription activation. Elf4 regulates the expression of genes involved in the transition from G0 to G1 phase of the cell cycle. The transcription factor contributes significantly to T cell proliferation and NK cell maturation highlighting its role in adaptive and innate immune responses.

Pathways

The E74-like factor 4 fits importantly into the JAK-STAT and MAPK signaling pathways. These pathways are central to cellular responses to a broad range of cytokines and growth factors. In the JAK-STAT pathway Elf4 interacts with proteins such as STAT1 and STAT3 enhancing transcription of target genes involved in immune responses. Within the MAPK pathway its role involves modulation of cell cycle and survival signals emphasizing its regulatory influence in immune-related contexts.

Research links Elf4 to leukemias and immune system dysfunctions. Its dysregulation can lead to abnormal proliferation of hematopoietic cells contributing to diseases like acute myeloid leukemia. Elf4 interacts with related proteins such as FLT3 influencing leukemogenesis. Additionally alterations in Elf4 activity can impair immune cell function potentially leading to defective immune responses or autoimmune disorders. This connection makes Elf4 a potential target for therapeutic interventions in immune-related diseases.

Target data

Transcriptional activator that binds to DNA sequences containing the consensus 5'-WGGA-3'. Transactivates promoters of the hematopoietic growth factor genes CSF2, IL3, IL8, and of the bovine lysozyme gene. Acts synergistically with RUNX1 to transactivate the IL3 promoter (By similarity). Transactivates the PRF1 promoter in natural killer (NK) cells and CD8+ T cells (PubMed : 34326534). Plays a role in the development and function of NK and NK T-cells and in innate immunity. Controls the proliferation and homing of CD8+ T-cells via the Kruppel-like factors KLF4 and KLF2 (By similarity). Controls cell senescence in a p53-dependent manner. Can also promote cellular transformation through inhibition of the p16 pathway. Is a transcriptional regulator of inflammation, controlling T-helper 17 (Th17) cells and macrophage inflammatory responses. Required for sustained transcription of anti-inflammatory genes, including IL1RN (PubMed : 34326534, PubMed : 35266071). Is a negative regulator of pro-inflammatory cytokines expression including IL17A, IL1B, IL6, TNFA and CXCL1 (PubMed : 34326534, PubMed : 35266071). Down-regulates expression of TREM1, a cell surface receptor involved in the amplification of inflammatory responses (By similarity) (PubMed : 34326534, PubMed : 35266071).
