ELL also known as Eleven-nineteen lysine-rich leukemia is a transcription elongation factor. It has a molecular mass of approximately 69 kDa. This protein facilitates the process of transcriptional elongation by RNA polymerase II. ELL is widely expressed in various tissues with notable expression in the immune and hematopoietic systems. It interacts directly with RNA polymerase II and increases the transcriptional output by suppressing pausing of the polymerase on the DNA template.
Biological function summary

ELL enhances transcription processes which are fundamental to gene expression regulation. It can form complexes with other proteins such as the ELL-associated factor (EAF) proteins which aids in its function. These complexes modulate transcriptional elongation by coordinating with transcription initiation factors and co-activators. This role is important for maintaining proper cellular functions and regulating gene expression patterns necessary for cell differentiation and proliferation.

Pathways

ELL is involved in significant cellular signaling like the transcription regulators pathway. It functions alongside other transcription regulation factors such as the p-TEFb (positive transcription elongation factor b) complex. It contributes to control transcriptional elongation cycle and maintaining proper transcriptional dynamics within cells. Through its pathway interactions ELL works with Cyclin T1 from the p-TEFb complex influencing cell cycle and development processes.

ELL has implications in leukemia especially acute myeloid leukemia (AML). Chromosomal translocations that involve ELL result in fusion proteins that disrupt normal transcription processes leading to oncogenesis. ELL fusion proteins may interact with MLL (mixed-lineage leukemia) proteins in these conditions altering regulation of target genes and contributing to leukemia pathogenesis. ELL's malfunction also connects to pathological states involving transcriptional dysregulation impacting cellular growth and differentiation pathways linked with cancerous transformations.

Elongation factor component of the super elongation complex (SEC), a complex required to increase the catalytic rate of RNA polymerase II transcription by suppressing transient pausing by the polymerase at multiple sites along the DNA. Elongation factor component of the little elongation complex (LEC), a complex required to regulate small nuclear RNA (snRNA) gene transcription by RNA polymerase II and III (PubMed : 22195968, PubMed : 23932780). Specifically required for stimulating the elongation step of RNA polymerase II- and III-dependent snRNA gene transcription (PubMed : 23932780). ELL also plays an early role before its assembly into in the SEC complex by stabilizing RNA polymerase II recruitment/initiation and entry into the pause site.
