ELMO1 and ELMO2 are proteins that play a role in intracellular signaling and cytoskeletal organization. ELMO1 is approximately 82 kDa in mass while ELMO2 is slightly smaller at around 80 kDa. These proteins are expressed in various tissues including the brain heart and lymphoid tissues. ELMO1 and ELMO2 interact with Dock180 a guanine nucleotide exchange factor to modulate cell morphology and migration by controlling actin cytoskeleton dynamics.
Biological function summary

ELMO1 and ELMO2 facilitate the regulation of cell migration phagocytosis and apoptosis. They are parts of a larger protein complex involving Dock180 and Rac1 contributing to the reorganization of the actin cytoskeleton. The ELMO-Dock180 complex acts as a critical mediator for the Rac signaling pathway affecting how cells respond to external signals that guide movement and cell survival.

Pathways

ELMO1 and ELMO2 participate in signaling pathways like the Rac1 pathway and the PI3K/Akt pathway. Through interactions with Rac1 these proteins regulate cytoskeletal rearrangements essential for various cellular processes including movement and immune cell function. ELMO1 and ELMO2 also connect with PI3K influencing the PI3K/Akt signaling cascade that is involved in cell growth and survival.

ELMO1 and ELMO2 have implications in cancer and autoimmune diseases. Studies link ELMO1 to increased susceptibility to systemic lupus erythematosus. Altered expression of ELMO2 is associated with cancer progression highlighting its potential as a therapeutic target. In cancer the interaction between ELMO proteins and Rac1 can enhance cell migration contributing to metastasis.
