Elp4 also known as Elongator complex protein 4 has an important role in the transcription process within cells. Its molecular weight is approximately 46 kDa. Elp4 is expressed in multiple tissues throughout the body with higher presence observed in the brain. It works as a component of the larger Elongator complex collaborating with other proteins such as Elp1 Elp2 Elp3 Elp5 and Elp6.
Biological function summary

Elongator complex proteins contribute to various cellular functions. Elp4 plays an important role in facilitating the modification of tRNA molecules necessary for efficient protein translation. It acts as part of the Elongator complex which participates in transcriptional elongation impacting processes like acetylation of histone 3 lysine 14. Elp4's activity in these processes ensures the proper progression and regulation of gene expression.

Pathways

Elp4 integrates into several significant cellular pathways. Notably it is active in the transcriptional elongation pathway an important process for proper gene expression. It also interacts with the oxidative stress response pathway aiding cells to manage stress. Within these pathways Elp4 collaborates closely with other elements of the Elongator complex ensuring synchronized functioning in transcription and stress response.

Research indicates that mutations or dysfunctions in Elp4 can contribute to several neurological conditions particularly juvenile-onset dystonia. This connection links Elp4 indirectly with other proteins involved in neural function such as dopamine receptor pathways. Additionally Elp4 is implicated in neurodevelopmental disorders like Rolandic epilepsy where its interaction with other components in the neuronal signaling may be disrupted.

Component of the elongator complex which is required for multiple tRNA modifications, including mcm5U (5-methoxycarbonylmethyl uridine), mcm5s2U (5-methoxycarbonylmethyl-2-thiouridine), and ncm5U (5-carbamoylmethyl uridine) (PubMed : 29332244). The elongator complex catalyzes the formation of carboxymethyluridine in the wobble base at position 34 in tRNAs (PubMed : 29332244).
