Supplementary information

EMAP II also known as AIMP1 (Aminoacyl tRNA Synthetase Complex-Interacting Multifunctional Protein 1) is a protein with a mass of approximately 34 kDa. Mechanically it modulates various cellular processes including apoptosis and inflammation. This protein is expressed in several tissues with higher levels seen in the immune cells the lung and the heart. AIMP1 is part of the wider group of proteins involved in the multi-synthetase complex which ensures efficient protein synthesis.
Biological function summary

AIMP1 functions beyond its role in the multi-synthetase complex to affect cellular proliferation and differentiation. It acts as a cytokine that can influence immune responses. When secreted AIMP1 shows pro-inflammatory activity targeting endothelial cells macrophages and fibroblasts. It is important for regulating the immune system's response to stress and external stimuli thereby bridging cellular activities with immune signaling networks.

Pathways

AIMP1 integrates into both the inflammatory response pathway and apoptosis signaling. This protein interacts with TNF-alpha and IL-6 which are important players in inflammation. AIMP1's activity influences ERK and JNK pathways contributing to the regulation of apoptosis and survival of immune cells. Its relation with these pathways highlights its role as a mediator that can shift cellular balance towards survival or programmed cell death based on external cues.

AIMP1 has connections to cancer and autoimmune diseases. Studies show increased levels of AIMP1 in tumors linking it to cancer progression especially in breast cancer where it interacts with proteins like p53. Furthermore AIMP1's involvement in autoimmune disorders such as rheumatoid arthritis connects it with TNF-alpha an important inflammatory mediator. This connection suggests AIMP1's function as a potential biomarker and target for therapeutic interventions in such conditions.

Target data

Non-catalytic component of the multisynthase complex. Stimulates the catalytic activity of cytoplasmic arginyl-tRNA synthase (PubMed : 10358004). Binds tRNA. Possesses inflammatory cytokine activity (PubMed : 11306575). Negatively regulates TGF-beta signaling through stabilization of SMURF2 by binding to SMURF2 and inhibiting its SMAD7-mediated degradation (By similarity). Involved in glucose homeostasis through induction of glucagon secretion at low glucose levels (By similarity). Promotes dermal fibroblast proliferation and wound repair (PubMed : 16472771). Regulates KDELR1-mediated retention of HSP90B1/gp96 in the endoplasmic reticulum (By similarity). Plays a role in angiogenesis by inducing endothelial cell migration at low concentrations and endothelial cell apoptosis at high concentrations (PubMed : 12237313). Induces maturation of dendritic cells and monocyte cell adhesion (PubMed : 11818442). Modulates endothelial cell responses by degrading HIF-1A through interaction with PSMA7 (PubMed : 19362550).
