Cell surface receptor that binds to the chondroitin sulfate moiety of glycosaminoglycan chains and promotes cell attachment. Promotes granulocyte chemotaxis, degranulation and adhesion. In macrophages, promotes the release of inflammatory cytokines, including IL8 and TNF. Signals probably through G-proteins. Is a regulator of mast cell degranulation (PubMed:26841242).

Activity summary

EMR2 also known as epidermal growth factor-like module receptor 2 or CD312 is a member of the class B G protein-coupled receptor family. It is a large receptor with an approximate mass of 220 kDa. EMR2 is mainly expressed on leukocytes especially myeloid cells like monocytes and macrophages. EMR2 consists of an extensive extracellular domain that is involved in ligand binding influencing cell adhesion and signaling processes.

Biological function summary

EMR2 plays a role in the immune system by facilitating cell-cell interactions and guiding leukocyte trafficking. It forms part of a receptor complex that includes other G protein-coupled receptors. The receptor is essential in the regulation of inflammatory responses helping in the recognition and response to pathogens or tissue damage. EMR2's function contributes to the activation and migration of immune cells to sites of infection or injury.

Pathways

EMR2 interacts with intracellular signaling cascades such as the MAPK and PI3K pathways that manage cell proliferation survival and immune responses. Through these pathways EMR2 connects to proteins such as Akt and ERK facilitating downstream signaling mechanisms important for immune cell activation. The integration of EMR2 in these pathways positions it as an important player in cellular responses to external stimuli.

Associated diseases and disorders

Aberrations in EMR2 function or expression relate to chronic inflammatory conditions and autoimmune disorders. Conditions such as rheumatoid arthritis and inflammatory bowel disease show altered EMR2 signaling indicating its role in the pathology of these diseases. In the context of these disorders other proteins like TNF-alpha and IL-6 often interact with EMR2 linking it to pro-inflammatory pathways and the exacerbation of disease symptoms.

    Interpolated concentration of native EMR2 was measured at different sample concentrations. Undiluted sample is as follows: U-937 cell extract (1 mg/mL). The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- standard deviation, n=2). Sample dilutions are made in 1% bovine serum albumin (BSA). Data represented here are indicative of assay working using example of biological sample. However, we do not guarantee same sensitivity level due to biological variation.
