Supplementary information

The Emx1 protein also known as Empty Spiracles Homeobox 1 acts as a transcription factor involved in the regulation of gene expression. Emx1 has a mass of approximately 29 kDa. It is expressed predominantly in the developing central nervous system particularly in the cerebral cortex and olfactory bulbs. During brain development Emx1 plays an important role in controlling the patterning and positioning of neurons.
Biological function summary

This transcription factor regulates the expression of various genes essential for brain structure organization and neuron differentiation. Emx1 often operates alongside other homeodomain proteins as part of transcriptional regulatory complexes to influence cortical development. These complexes guide the regional specification and morphological differentiation making Emx1 significant for organizing the structure of the brain.

Pathways

Emx1 is intricately involved in neuronal specification and differentiation pathways. It operates within the Wnt signaling pathway collaborating with proteins like Lhx2 to regulate neural progenitor cell fate and proliferation. The integration of Emx1 in these pathways ensures proper neuronal development and cerebral cortex maturation essential for brain function.

Mutations or dysregulation of Emx1 have associations with neurodevelopmental disorders such as autism spectrum disorder (ASD) and epilepsy. In these conditions Emx1's interaction with other transcription factors like Pax6 may contribute to abnormal neural development or cortical malformations highlighting its importance in maintaining normal brain function.

Target data

Transcription factor, which in cooperation with EMX2, acts to generate the boundary between the roof and archipallium in the developing brain. May function in combinations with OTX1/2 to specify cell fates in the developing central nervous system.
