EMX2 also known as Empty Spiracles Homeobox 2 functions as a homeobox transcription factor. This protein has a mass of approximately 33 kilodaltons. It is mainly expressed in the developing brain where it plays an important role in brain morphogenesis and neuronal development. EMX2 also presents in other tissues such as the kidney and reproductive organs where it contributes to organogenesis.
Biological function summary

The EMX2 protein influences developmental processes by regulating gene expression essential for cellular differentiation and tissue development. It does not belong to a larger complex but interacts with various transcription factors to exert its regulatory functions. EMX2 modulates important aspects of brain development guiding the patterning and regional specification within the cerebral cortex.

Pathways

EMX2 plays an influential role in the Wnt signaling pathway which is key to developmental processes like cell fate determination proliferation and migration. Additionally EMX2 is involved in the regulation of neural lineage specification through its interaction with other proteins such as PAX6. Together these proteins create a regulatory network that controls neural precursor cell proliferation and differentiation.

EMX2 has implications in neurological disorders. Abnormal expression or mutations in the EMX2 gene can contribute to developmental brain disorders such as schizencephaly which involves abnormal clefts or deep grooves in the cerebral hemispheres. The protein also links to cancer pathways notably in gliomas where dysregulation of EMX2 expression can affect tumor progression. Connections between EMX2 and proteins like GLI1 in the Glioma signaling pathway provide insights into its role in oncogenesis.

Target data

Transcription factor, which in cooperation with EMX1, acts to generate the boundary between the roof and archipallium in the developing brain. May function in combination with OTX1/2 to specify cell fates in the developing central nervous system. In the inner ear, it controls the distribution of GPR156 at hair cell boundaries, and regulates the organization of stereociliary bundles in opposite orientations across the line of polarity reversal (LPR).
