The target EN1 also known as Engrailed 1 is a homeobox-containing transcription factor involved in developmental processes. EN1 protein weighs approximately 40 kDa and is expressed primarily in the developing brain especially in the midbrain and hindbrain regions during embryonic development. It also appears in the skeletal and nervous systems where it regulates specific gene expression patterns. The protein sometimes referred to as EN1 protein plays a significant role in tissue specification and morphogenesis during early developmental stages.
Biological function summary

The Engrailed 1 protein influences pattern formation by controlling the transcription of key developmental genes. Part of several transcriptional complexes it binds to DNA sequences aiding in the specification of positional identities during embryogenesis. EN1 also has significant functions in the maintenance of cell fates and is essential for the differentiation of numerous cell types. Through these processes EN1 ensures proper segmental development and organogenesis.

It also interacts with other transcription factors such as Pax2 in the regulation of pathways linked to neural development.

EN1 has connections to conditions like Parkinson's disease and certain congenital disorders like cerebellar hypoplasia. The dysregulation or mutation of EN1 influences the onset and progression of these diseases. It relates to the serotoninergic and dopaminergic systems reflecting its importance in maintaining normal neurotransmitter functions. In Parkinson's the interaction of EN1 with proteins such as α-synuclein demonstrates its potential impact on neurodegeneration and neuron survival.
