Lanes 1 - 4 : Merged signal (red and green). Green - ab76122 observed at 29 kDa. Red - loading control, ab7291, observed at 52 kDa.

ab76122 was shown to specifically react with Endo G when Endo G knockout samples were used. Wild-type and Endo G knockout samples were subjected to SDS-PAGE. ab76122 and ab7291 (loading control to alpha tubulin) were diluted 1/2000 and 1/10000 respectively and incubated overnight at 4°C. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD)preadsorbed (ab216776) secondary antibodies at 1/10000 dilution for 1 h at room temperature before imaging.

Endonuclease G (Endo G) is an important mitochondrial nuclease known for its role in DNA and RNA metabolism. Endo G is also referred to as endoglycosidase S and has a molecular mass of approximately 28 kDa. This protein is expressed mainly in the mitochondria but can translocate to the nucleus under certain conditions. It functions by cleaving DNA and RNA which facilitates the processing of nucleic acids in the cell. Endo G exhibits endonuclease activity making it a significant factor in nucleic acid regulation.
Biological function summary

This target plays an essential role in mitochondrial DNA fragmentation during apoptosis. Endo G operates independently of caspases highlighting its role as a non-canonical mediator of apoptosis. It becomes active upon cellular stress usually through the release from the mitochondria where it functions as part of a larger apoptotic complex. This characteristic grants it an important function in programmed cell death mechanisms.

Pathways

Endo G integrates into apoptosis and mitochondrial biogenesis. This protein participates in the intrinsic pathway of apoptosis collaborating with other proteins like apoptosis-inducing factor (AIF) to execute cell death. Endonuclease that preferentially catalyzes the cleavage of double-stranded 5-hydroxymethylcytosine (5hmC)-modified DNA (PubMed : 25355512). The 5hmC-modified nucleotide does not increase the binding affinity, but instead increases the efficiency of cutting and specifies the site of cleavage for the modified DNAs (By similarity). Shows significantly higher affinity for four-stranded Holliday junction over duplex and single-stranded DNAs (By similarity). Promotes conservative recombination when the DNA is 5hmC-modified (PubMed : 25355512). Promotes autophagy through the suppression of mTOR by its phosphorylation-mediated interaction with YWHAG and its endonuclease activity-mediated DNA damage response (PubMed : 33473107). GSK3-beta mediated phosphorylation of ENDOG enhances its interaction with YWHAG, leading to the release of TSC2 and PIK3C3 from YWHAG resulting in mTOR pathway suppression and autophagy initiation (PubMed : 33473107). Promotes cleavage of mtDNA in response to oxidative and nitrosative stress, in turn inducing compensatory mtDNA replication (PubMed : 29719607).
