ENDOGL1/ENGL was immunoprecipitated from 1mg of HUVEC (Human umbilical vein endothelial cell line) whole cell lysate with ab206300 at 1/40 dilution. Western blot was performed from the immunoprecipitate using ab206300 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366), was used for detection at 1/10000 dilution.

Lane 1 : HUVEC whole cell lysate 10ug (Input). Lane 2 : ab206300 IP in HUVEC whole cell lysate. Lane 3 : Rabbit monoclonal IgG (ab172730) instead of ab206300 in HUVEC whole cell lysate.

Blocking and dilution buffer and concentration : 5% NFDM/TBST.

Exposure time : 10 seconds.

ENDOGL1 also known as Endonuclease G-Like 1 (ENGL) serves as an important enzyme in the cell that cleaves RNA. Its mass is approximately 35 kilodaltons. ENDOGL1 expresses mainly in the mitochondria where it acts closely with other mitochondrial components. This expression is also observed in various tissues throughout the body indicating a broad functional scope. As a ribonuclease it participates in the degradation of RNA which affects the processing and maturation of mitochondrial RNA.
Biological function summary

The role of ENDOGL1 centers around its participation in the maintenance of mitochondrial RNA stability. ENDOGL1 functions within a larger mitochondrial complex contributing to RNA turnover and regulation. By engaging directly with the RNA it plays a significant role in controlling and managing RNA fragments within the mitochondria essential for normal mitochondrial operations. This function ensures the smooth synthesis of the components required for mitochondrial respiration and energy production.

ENDOGL1 integrates into mitochondrial RNA decay and processing pathways working alongside key proteins like POLRMT and TFAM involved in mitochondrial transcription and replication. ENDOGL1 influences the integrity and efficiency of these pathways by controlling RNA fragment degradation. Its activity in these pathways supports mitochondrial homeostasis and helps manage the mitochondrial gene expression thereby facilitating various energy production processes such as oxidative phosphorylation.
