The epitope recognized by the UMB-8-37-1 monoclonal contains phosphorylation sites that affect signal in Western blot after treatment with endothelin. Phosphatase treatment may be required prior to Western blot in order to improve signal (PubMed ID 25708416).

The Endothelin A Receptor (ET-A) also known as endothelin receptor type A is a G protein-coupled receptor responsible for mediating physiological responses to the endothelin family of peptides. It has a molecular weight of approximately 48 kDa. ET-A is expressed in various tissues particularly in vascular smooth muscle cells and certain neural tissues including those resembling ganglioid structures. The engagement of the receptor by endothelin leads to a cascade of intracellular events that result in vasoconstriction and increased blood pressure.
ET-A functions by binding endothelin peptides specifically endothelin 1 initiating a series of reactions essential for vascular homeostasis. It forms a part of receptor signaling complexes that propagate downstream signals influencing cardiovascular tone and cell proliferation. ET-A holds an important role in modulating secretion of hormones and controlling the nervous system by interacting with signaling molecules.
ET-A is heavily involved in the endothelin signaling pathway which interacts with the renin-angiotensin system. Through these pathways it regulates blood pressure and vascular tone. Endothelin receptor type A interfaces with proteins such as angiotensin-converting enzyme influencing vasoconstriction activities and impacting cardiovascular health. These interactions highlight ET-A's important regulatory role in maintaining vascular and cardiac function.
ET-A aligns with conditions like hypertension and pulmonary arterial hypertension. The receptor's overactivity results in elevated vasoconstrictor tone therefore contributing to the mentioned disorders. It associates with proteins like the angiotensin II receptor in these contexts impacting pathological states where vascular balance is disrupted. Understanding the modulation of ET-A offers potential therapeutic strategies for these cardiovascular disorders.
