Epac2 also known as exchange protein directly activated by cAMP 2 functions as a guanine nucleotide exchange factor (GEF) for the small GTPases Rap1 and Rap2. Epac2 weighs approximately 110 kDa and is expressed in multiple tissues with high expression in the brain and pancreas. It responds to the second messenger cAMP acting as a critical link between G protein-coupled receptors (GPCRs) and downstream signal transduction pathways. Epac2 has two important domains: the regulatory cAMP-binding domain and the catalytic CDC25 homology domain allowing it to operate as a multi-functional signaling node.
Biological function summary

Epac2 modulates intracellular signaling processes by mediating the activation of small GTPases. This activation influences cell adhesion cell junction formation and insulin secretion. Epac2 does not function in isolation; it forms a complex with other signaling molecules impacting various cellular processes. In the brain Epac2 also plays a significant role in synaptic plasticity and memory formation integrating signals that modulate neuronal activity.

Pathways

Epac2 integrates into key signaling cascades such as the cAMP signaling pathway and the MAPK/ERK pathway. It participates in the cAMP signaling pathway by facilitating conversion of cAMP to active GTP-bound Rap proteins. In doing so Epac2 interacts closely with proteins like protein kinase A (PKA) and Rap1. Each component plays a distinct role in transducing extracellular signals into cellular responses promoting a coordinated cellular activity profile.

Research connects Epac2 to conditions such as type 2 diabetes and autism spectrum disorders (ASD). In type 2 diabetes Epac2 influences insulin secretion and its dysregulation may be linked to impaired glucose homeostasis. In ASD altered Epac2 signaling impacts neuronal communication and plasticity. Beyond this Epac2's interaction with other regulatory proteins like Rap1 plays a role in these disease contexts by affecting cellular functions critical for metabolic and neurological health.
