EPH receptors A3 A4 and A5 which are also known as EPHA3 EPHA4 and EPHA5 are part of the receptor tyrosine kinase family. These proteins have a mass around 100-130 kDa. They predominantly express in neural tissues but one can also find them in certain epithelial and mesenchymal tissues. Functionally they play a role in cell-cell communication by binding with ephrin ligands which leads to kinase activation and downstream signaling cascades.
Biological function summary

EPH receptors A3 A4 and A5 are essential for mediating a range of cellular processes including cell migration axon guidance and tissue boundary formation. They do not act in isolation but often form complexes with ephrin ligands such as ephrin-A1 and ephrin-A5. These interactions are critical in developmental processes and maintaining cellular architecture. By influencing cell positioning and movement they significantly contribute to nervous system development and vascular patterning.

Pathways

EPH A3 A4 and A5 integrate into the ephrin signaling pathway and the integrin-mediated signaling pathway. In the ephrin signaling pathway they work in tandem with related proteins such as EPHA2 and EPHB receptors to modulate cellular dynamics. These pathways dictate cytoskeletal organization influencing cellular adhesion and motility impacting a variety of physiological processes including angiogenesis and nerve development.

Altered functions of EPH A3 A4 and A5 associate with cancer and neurodegenerative diseases. In cancer especially glioblastoma and melanoma their overexpression or aberrant activity can lead to enhanced cell proliferation and metastasis partially through interactions with proteins like EPHA2. In neurodegenerative conditions such as Alzheimer's disease dysregulated EPH receptor signaling may contribute to neuronal death and cognitive impairment connected with proteins involved in tau pathologies.

Target data

Receptor tyrosine kinase which binds promiscuously GPI-anchored ephrin-A family ligands residing on adjacent cells, leading to contact-dependent bidirectional signaling into neighboring cells. The signaling pathway downstream of the receptor is referred to as forward signaling while the signaling pathway downstream of the ephrin ligand is referred to as reverse signaling. Among GPI-anchored ephrin-A ligands, EFNA5 most probably constitutes the cognate/functional ligand for EPHA5. Functions as an axon guidance molecule during development and may be involved in the development of the retinotectal, entorhino-hippocampal and hippocamposeptal pathways. Together with EFNA5 plays also a role in synaptic plasticity in adult brain through regulation of synaptogenesis. In addition to its function in the nervous system, the interaction of EPHA5 with EFNA5 mediates communication between pancreatic islet cells to regulate glucose-stimulated insulin secretion (By similarity).
