Ephexin-1 also referred to as NGEF (neuronal guanine nucleotide exchange factor) acts as a guanine nucleotide exchange factor (GEF) that specifically activates Rho GTPases by promoting the exchange of GDP for GTP. Ephexin-1 has an approximate mass of 70 kDa. Its expression is widely observed in various tissues with significant levels in the nervous system where it plays important roles in neural development and function.
Biological function summary

Ephexin-1 plays a role in cytoskeletal reorganization by modulating the activity of Rho GTPases which are critical regulators of actin dynamics. It is part of molecular complexes that help control axon guidance and synapse formation in neurons. Through these functions Ephexin-1 influences cell shape motility and growth.

Pathways

Ephexin-1 interacts with neural pathways such as the ephrin signaling pathway. It acts in conjunction with proteins like Eph receptors which are important for cell-cell communication and guidance during developmental processes. Target data

Acts as a guanine nucleotide exchange factor (GEF) which differentially activates the GTPases RHOA, RAC1 and CDC42. Plays a role in axon guidance regulating ephrin-induced growth cone collapse and dendritic spine morphogenesis. Upon activation by ephrin through EPHA4, the GEF activity switches toward RHOA resulting in its activation. Activated RHOA promotes cone retraction at the expense of RAC1- and CDC42-stimulated growth cone extension (By similarity).
