The human prostate cancer section was performed by LeicaDS9800 (Bond™ Polymer Refine Detection). The sections were using Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0, epitope retrieval solution 2) for 20 mins, then labelling ab284823 at 1/4000 dilution for 30 mins at room temperature. Counterstained with hematoxylin. The immunostaining was performed on a Leica Biosystems BOND® RX instrument.

EPS15 also known as Epidermal Growth Factor Receptor Substrate 15 is a critical adaptor protein involved in the process of endocytosis. The protein has a molecular mass of approximately 105 kDa and is highly expressed in various tissues particularly in the nervous system. EPS15 acts as an important player in clathrin-mediated endocytosis by interacting with components of endocytic machinery such as clathrin and AP2 adaptor complexes. It also contains multiple EH domains which facilitate protein-protein interactions needed for the internalization of receptors.
Biological function summary

The EPS15 protein contributes to the regulation of cell surface receptors and is involved in the recycling of these receptors back to the membrane or directing them to lysosomal degradation. This protein partakes in the formation of the clathrin-coated pits which are essential for the invagination of the membrane in the endocytic process. EPS15 works within a complex that aids in trafficking signals within cells allowing cells to maintain homeostasis by dynamically responding to external stimuli.

Pathways

EPS15 significantly influences the endocytosis and EGFR signaling pathways. The protein facilitates the downregulation of receptor tyrosine kinase signaling by mediating the internalization and recycling of the epidermal growth factor receptor (EGFR). EPS15 also interacts with other proteins such as Grb2 and Epsin which are integral to these pathways. The relationship between EPS15 and these pathways highlights its importance in cell communication and proliferation control.

Dysfunction of EPS15 links to cancer and neurological disorders. Abnormalities in EPS15 expression or function can contribute to cancer progression by disrupting EGFR signaling leading to unregulated cell growth and division. In neurological contexts altered EPS15 expression may affect synaptic transmission and faulty receptor trafficking potentially playing a role in neurodegenerative diseases. Involved in the internalization of ligand-inducible receptors of the receptor tyrosine kinase (RTK) type, in particular EGFR. Plays a role in the assembly of clathrin-coated pits (CCPs). Acts as a clathrin adapter required for post-Golgi trafficking. Seems to be involved in CCPs maturation including invagination or budding. Involved in endocytosis of integrin beta-1 (ITGB1) and transferrin receptor (TFR); internalization of ITGB1 as DAB2-dependent cargo but not TFR seems to require association with DAB2.
