Lanes 1 - 3 : Merged signal (red and green). Green - ab32121 observed at 185 kDa. Red - loading control, ab9484, observed at 37 kDa.

ab32121 was shown to specifically recognize ERBB3 in wild-type HAP1 cells along with additional cross-reactive bands. No band was observed when knock out samples were examined. Wild-type and ERBB3 knockout samples were subjected to SDS-PAGE. ab32121 and ab18058 (Mouse anti Vinculin loading control) were incubated overnight at 4°C at 1/500 dilution and 1/20,000 dilution respectively. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed (ab216776) secondary antibodies at 1/20,000 dilution for 1 hour at room temperature before imaging.

The ErbB3 protein also known as HER3 is a member of the epidermal growth factor receptor (EGFR) family. It belongs to the ErbB receptor tyrosine kinase family but lacks intrinsic kinase activity. The HER3 protein has a molecular weight of approximately 180 kDa. You will find HER3 expressed in various tissues with high expression in the epithelial tissues where it plays a role in normal cellular processes. HER3 interacts with multiple ligands facilitating cellular communication and growth regulation.
Biological function summary

HER3 is essential for activating important signaling pathways that regulate proliferation differentiation and survival. The ErbB3 protein forms heterodimers with other ErbB family members such as ErbB2 (HER2) which is necessary for its signaling capability. The HER3 protein is abundant in heart and nervous tissues indicating its role in cardiovascular and neurological functions.
