This IP data was generated using the same anti-ErbB4 antibody clone, E200, in a different buffer formulation (cat# ab32375).
Lane 1 (input) : HEK-293 (human embryonic kidney epithelial cell) whole cell lysate, 10μg
Lane 2 (+) : HEK-293 whole cell lysate
Lane 3 (-) : Rabbit monoclonal IgG (ab172730) instead of ab32375 in HEK-293 whole cell lysate
ab32375 Immunoprecipitating ErbB 4 in HEK-293 whole cell lysate. Capture antibody was used at a 1 : 50 dilution (2μg in 0.35mg lysates). For western blotting, primary antibody used as ab32375 at 1 : 500 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366), was used for detection at 1/1000 dilution. The lower band at around 75kDa should be proteolysis fragment based on the literature. (PMID : 9362517)

This data was developed using the same antibody clone in a different buffer formulation (ab32375).
Western blot : Anti-ErbB4 / HER4 antibody [E200] ab32375 staining at 1/2000 dilution, shown in green; Mouse anti-CANX (ab238078) loading control staining at 1/20,000 dilution, shown in magenta. A band was observed at 171 kDa in T-47D cell lysates with no signal observed at this size in A549 cell line. To generate this image, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 100 % LICOR in TBS-0.1 % Tween® 20 (TBS-T) before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged. Secondary antibodies used were Goat anti-Rabbit 800CW & Goat anti-Mouse 680RD at 1/20,000 dilution.

ErbB4 has several roles including the regulation of cell proliferation differentiation migration and development. It functions as part of a receptor complex initiating a cascade of intracellular signaling that impacts various cellular functions. ErbB4 ELISA assays often help quantify interactions and activities of the receptor providing insights into its broader functionalities. The ErbB4 protein also contributes to neuronal development and the maintenance of cardiac tissue integrity.
Pathways
ErbB4 participates in the PI3K/AKT pathway and the MAPK/ERK pathway essential for cell survival and proliferation.
