ERC2 also known as ELKS/Rab6-interacting/CAST family member 2 is part of the ELKS protein family. It weighs approximately 120 kDa. ERC2 is abundantly expressed in the brain and other neural tissues. This protein is mainly located in the presynaptic active zone of neurons supporting its role in synaptic function by helping to organize and maintain the structure necessary for neurotransmitter release.
ERC2 plays an important role in synaptic vesicle exocytosis. It supports the assembly and stabilization of the presynaptic machinery which includes interacting with other synaptic proteins like RIM and Munc13 to facilitate neurotransmitter release. ERC2 is an integral part of a complex with other active zone proteins contributing to efficient signal transfer across synapses by enabling rapid and reliable neurotransmitter release.
ERC2 operates within the synaptic vesicle cycle and the neurotransmitter release pathway. It engages closely with proteins such as RIM1 and Liprin-α which are also essential for organizing the presynaptic grid. These interactions are fundamental to maintaining the synaptic active zone's architecture therefore promoting precise signal transduction in neural communication.
ERC2 has been linked to neurological disorders such as epilepsy and autism spectrum disorders. Alterations in its expression or function can disrupt synaptic transmission which may contribute to these conditions. ERC2 interacts with other proteins involved in these disorders like Munc13 which is known to affect synaptic plasticity and neurotransmitter release.
