ERG2 also known as sterol isomerase or C-8 sterol isomerase is an enzyme in the ergosterol biosynthesis pathway important for sterol production. This protein has a mass of approximately 25 kDa. ERG2 is found mainly in the endoplasmic reticulum and is highly expressed in yeast and fungi. ERG2's primary role involves catalyzing the isomerization of sterols an important step in ergosterol synthesis from which the eukaryotic cell membranes gain structural integrity.
Biological function summary

Sterol isomerase activity is significant in maintaining membrane fluidity and sterol homeostasis. ERG2 functions as part of the larger ergosterol biosynthesis enzyme complex. The integrity and function of the cellular membrane depend on the proper functioning of this complex with ERG2 being one of the essential modulators within it.

Pathways

ERG2 operates within the ergosterol biosynthesis pathway which is part of a larger lipid metabolic pathway important for maintaining cellular functions. This pathway includes several other enzymes like ERG3 and ERG6 which work consecutively with ERG2. Disruptions in this pathway can lead to cell membrane instability and defects in organismal growth regulation.

Defects or inhibition of ERG2 can result in problems like ergosterol biosynthesis disorders primarily in fungi making it an important target for antifungal drugs. Conditions related to ERG2 also involve cholesterol synthesis in broader biological systems affected through its interactions with proteins like ERG11. Disorders arising from ergosterol pathway disruptions may affect cellular proliferation and development leading to broader systemic implications in pathogenic fungi.

Pore-forming (alpha) subunit of voltage-gated inwardly rectifying potassium channel. Characterized by unusual gating kinetics by producing relatively small outward currents during membrane depolarization and large inward currents during subsequent repolarization which reflect a rapid inactivation during depolarization and quick recovery from inactivation but slow deactivation (closing) during repolarization. Activates even more slowly than KCNH2.
