ab102010 staining ERM / Etv5 in HeLa cells by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with 4% paraformaldehyde at room temperature for 15 minutes. Samples were incubated with primary antibody (1/500) and cytoskeleton stained red with Phalloidin.

Lanes 1 - 2 : Merged signal (red and green). Green - ab102010 observed at 58 kDa. Red - loading control ab8245 (Mouse anti-GAPDH antibody [6C5]) observed at 37 kDa.

ab102010 was shown to react with ERM / Etv5 in Western blot. Membranes were blocked in 3 % milk in TBS-T (0.1 % Tween®) before incubation with ab102010 and ab8245 (Mouse anti-GAPDH antibody [6C5]) overnight at 4°C at a 1 in 500 dilution and a 1 in 20000 dilution respectively. Blots were incubated with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed (ab216776) secondary antibodies at 1 in 20000 dilution for 1 h at room temperature before imaging.

The ERM (Etv5) protein also known as ERM products or ERM Boston in some contexts functions as a transcription factor within the ETS family of proteins. This protein has a molecular weight of approximately 57 kDa. Expressed in a variety of tissues including the brain and reproductive organs Etv5 plays an important role in regulating gene expression by binding to specific DNA sequences. Its activities involve promoting transcription of genes vital for development and cellular differentiation.
Biological function summary

The ERM (Etv5) protein influences numerous developmental processes and cellular states. It participates in the MAP kinase signaling cascade where it facilitates responses to growth factors. By serving as a component in several regulatory complexes Etv5 modulates the expression of genes that control cell proliferation and survival. This activity is important for tissue development and maintenance especially in stem and progenitor cells.
