The ESGP protein short for Epithelial Sodium-Gated Protein serves as an important regulator in ion transport across epithelial cells. This protein approximately 90 kDa in molecular weight mainly expresses in the epithelial tissues of kidneys lungs and intestines. Alternative names for ESGP include ENaC (Epithelial Sodium Channel) and γ-ENaC which highlight its role as a subunit of the larger epithelial sodium channel complex.
Biological function summary

ESGP plays an integral role in the regulation of sodium balance impacting both fluid volume and blood pressure. As a part of the ENaC complex its activity in absorbing sodium ions from the lumen of epithelial surfaces significantly affects electrolyte homeostasis. Disregulation of ESGP function can cause a cascade of imbalances signaling its importance in maintaining cellular and systemic equilibrium.

Pathways

ESGP is pivotal in the renin-angiotensin-aldosterone system (RAAS) and the regulation of blood pressure pathways. In the RAAS pathway the ESGP associates with proteins like aldosterone-induced proteins to modulate sodium reabsorption. It also ties to proteins such as NEDD4 which influence its ubiquitination and therefore its activity within the pathway. This interaction demonstrates its central role in fluid regulation pathways.

ESGP has associations with conditions such as Liddle syndrome and cystic fibrosis. Liddle syndrome results from mutations that heighten the activity of ESGP leading to increased sodium retention and resulting hypertension. In cystic fibrosis aberrations in ESGP function contribute to abnormal salt transport across epithelial cells. In these conditions ESGP closely interacts with CFTR and other channel proteins that modulate ion transport highlighting its importance in maintaining ionic balance and disease mechanisms.
