ETAA1 also known as Ewing tumor-associated antigen 1 is a protein recognized for its role in DNA replication stress response. It is approximately 92 kDa in mass and ubiquitously expressed in various human tissues. ETAA1 functions at the mechanical level by activating the ATR (ataxia telangiectasia and Rad3-related) kinase an enzyme critical for maintaining genomic stability. This protein contains ATR activators that allow the phosphorylation of various substrates participating in DNA damage repair.
Biological function summary

ETAA1 plays a significant role in the cellular response to replication stress ensuring accurate DNA replication and repair. It does not function as a single entity but acts as part of the ATR activation complex which is involved in sensing and signaling DNA damage. By facilitating ATR activation ETAA1 helps coordinate cell cycle checkpoints and repair processes maintaining genomic integrity under stress conditions.

Pathways

ETAA1 integrates into the DNA damage response pathway and cellular cycle regulation. It operates closely with ATR within the DDR (DNA Damage Response) pathway sharing functional interactions with proteins like TopBP1 and Rad17 which also contribute to ATR activation. Together these proteins ensure the monitoring and repairing of DNA lesions therefore preventing the accumulation of mutations.

ETAA1 associates with cancer and developmental disorders linked to genomic instability. Its dysfunction or altered expression can disrupt the DNA damage response potentially leading to the accumulation of mutations seen in different cancers. The interaction of ETAA1 with ATR emphasizes this relationship as both proteins become pivotal in responding to DNA damage and maintaining cellular health.

Recruited to stalled replication forks via interaction with the RPA complex and directly stimulates ATR kinase activity independently of TOPBP1 (PubMed : 27723717, PubMed : 27723720, PubMed : 30139873).
