Supplementary information

ETNK2 also known as ethanolamine kinase 2 is an enzyme involved in the phospholipid biosynthesis pathway. This protein weighs approximately 44 kDa and is expressed in various tissues including the liver and brain. It plays an important role in the conversion of ethanolamine to phosphoethanolamine which is an important step in the production of phosphatidylethanolamine an important component of cell membranes.
Biological function summary

Ethanolamine kinase 2 facilitates the synthesis of phosphatidylethanolamine by functioning as part of the Kennedy pathway an essential process for membrane biogenesis and maintenance. Although not part of a known structural complex it functions closely with other metabolic enzymes to ensure efficient production of phospholipids. This enzyme contributes to maintaining proper cellular structures and signaling pathways by affecting membrane composition and dynamics.

Pathways

Ethanolamine kinase 2 significantly contributes to the CDP-ethanolamine pathway one of the two main routes for phosphatidylethanolamine biosynthesis. It works together with other enzymes like CTP:phosphoethanolamine cytidylyltransferase to regulate the availability of phosphatidylethanolamine. This pathway is important for maintaining cellular lipid homeostasis and supporting membrane-based cellular processes.

Ethanolamine kinase 2 has a connection to neurological conditions because of its significant role in phospholipid metabolism. Abnormalities in its function or expression can potentially lead to disorders such as metabolic syndrome and nonalcoholic fatty liver disease. In these contexts its activity links with other proteins involved in lipid metabolism such as phosphatidylethanolamine N-methyltransferase suggesting potential therapeutic targets within these pathways.
