Exportin 4 also known as XPO4 is a nuclear export receptor part of the karyopherin beta family. It has a molecular weight of approximately 130 kDa. This protein functions by mediating the transport of several cargo molecules from the nucleus to the cytoplasm engaging in the nucleocytoplasmic transport mechanism. Exportin 4 is expressed in various tissues but it shows a higher expression in tissues with active cell division such as the thymus. It specifically recognizes and binds to its cargo proteins in the presence of RanGTP facilitating their export through the nuclear pore complex.
Biological function summary

Exportin 4 plays an important role in the regulation of protein synthesis by exporting molecules involved in translation. It acts as part of a complex with RanGTP a small GTPase that provides the energy required for nuclear transport. This transport process is essential for maintaining cellular function and homeostasis by regulating the localization of proteins that are necessary for cell growth and division. The specificity of the cargo depends on its nuclear export signals which Exportin 4 recognizes with high affinity.

Pathways

Exportin 4 is integrated into the Ran pathway which is vital for nuclear import and export processes. In this pathway Exportin 4 transports proteins necessary for cell cycle progression and gene expression. A related protein in this pathway is RanBP1 which assists in the dissociation of the export complex in the cytoplasm. Exportin 4 also interacts with other proteins like CRM1 to coordinate the export of specific RNA molecules impacting the balance of nuclear and cytoplasmic activities in the cell.

Alterations in Exportin 4 function can influence cancer progression and neurodegenerative diseases. Aberrant export activity can lead to mislocalization of tumor suppressor proteins contributing to oncogenesis. In neurodegenerative conditions such as Alzheimer’s disease Exportin 4 dysregulation affects neuronal function partly due to its interaction with Tau protein which can aggregate abnormally in neurons. Understanding the dynamics of Exportin 4 can therefore provide insights into therapeutic targets for these diseases.

Target data

Mediates the nuclear export of proteins (cargos), such as EIF5A, SMAD3 and isoform M2 of PKM (PKM2) (PubMed : 10944119, PubMed : 16449645, PubMed : 26787900). In the nucleus binds cooperatively to its cargo and to the GTPase Ran in its active GTP-bound form. Docking of this trimeric complex to the nuclear pore complex (NPC) is mediated through binding to nucleoporins (PubMed : 10944119, PubMed : 16449645). Upon transit of a nuclear export complex into the cytoplasm, disassembling of the complex and hydrolysis of Ran-GTP to Ran-GDP (induced by RANBP1 and RANGAP1, respectively) cause release of the cargo from the export receptor (PubMed : 10944119, PubMed : 16449645). XPO4 then return to the nuclear compartment and mediate another round of transport (PubMed : 10944119, PubMed : 16449645). The directionality of nuclear export is thought to be conferred by an asymmetric distribution of the GTP- and GDP-bound forms of Ran between the cytoplasm and nucleus (PubMed : 10944119, PubMed : 16449645). Catalyzes the nuclear export of hypusinated EIF5A; a small cytoplasmic protein that enters nucleus and accumulates within nucleolus if not exported back by XPO4 (PubMed : 10944119). Specifically mediates nuclear export of isoform M2 of PKM (PKM2) following PKM2 deacetylation by SIRT6 (PubMed : 26787900). Also mediates the nuclear import of SOX transcription factors SRY and SOX2 (By similarity).
