Activity summary

F2RL3 also known as Protease-activated receptor 4 (PAR-4) is a G-protein coupled receptor with a molecular mass of approximately 42 kDa. This protein plays an important role in cellular responses to extracellular signals and is important for thrombin signaling in platelets. F2RL3 is expressed in a variety of tissues including the platelets vascular smooth muscle cells and endothelial cells. The receptor gets activated by proteolytic cleavage by thrombin and other proteases leading to a sequence of intracellular signaling events.

Biological function summary

F2RL3 is a significant component in coagulation and inflammatory response processes. The receptor participates in platelet activation and aggregation which contributes to blood clotting. F2RL3 forms part of a complex that includes other protease-activated receptors facilitating precise regulation of vascular function and homeostasis. The receptor also contributes to inflammation through interactions with immune cells impacting vascular and tissue inflammation.

Pathways

The function of F2RL3 integrates into the coagulation cascade and inflammatory signaling pathways. It interacts with other proteins like thrombin and F2RL1 (PAR-1) mediating platelet aggregation and extension of thrombotic processes. Through its action in pathways involving thrombin F2RL3 ensures effective hemostasis and also contributes to signaling pathways that mediate inflammation and tissue repair.

Associated diseases and disorders

F2RL3 has a relationship with thrombotic disorders and atherosclerosis. Abnormal activation or expression of F2RL3 can lead to excessive platelet activation increasing the risk of thrombosis. Furthermore its role in inflammatory pathways implies a connection to atherosclerosis a condition characterized by inflammation and plaque buildup in arteries. The receptor interacts with proteins such as PAR-1 during these disease processes which contributes to the pathophysiology of thrombotic and cardiovascular conditions.
