For immunofluorescence analysis, 70% confluent 3T3-L1 cells differentiated for 5 days were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton X-100 for 10 minutes, and blocked with 1% BSA for 1 hour at room temperature for detection of FABP4 using ab308110 at 2 µg/mL in 0.1% BSA and incubated for 3 hours at room temperature and then labeled with a Goat anti-Rabbit IgG (H+L) Secondary Antibody, Alexa Fluor® 488 conjugate at a dilution of 1/2000 for 45 minutes at room temperature (Panel a : green). Nuclei (Panel b : blue) were stained with DAPI F-actin (Panel c : red) was stained with Alexa Fluor® 555 Rhodamine Phalloidin at 1/300 dilution Panel d represents the merged image showing cytoplasmic localization. Western blot analysis of FABP4 in whole cell extracts of serum-starved 3T3 L1 cells treated with Insulin (100 ng/mL, 15 min) using ab308110 at a dilution of 1 µg/mL. Samples were detected using chemiluminescence (ECL). Results show a band at ~15 kDa.

Fatty acid-binding protein 4 (FABP4) also known as aP2 is a member of the fatty acid-binding protein family. It predominantly exists in adipocytes and macrophages serving an important role in lipid metabolism and transport. FABP4 facilitates the binding and transport of long-chain fatty acids and other hydrophobic molecules within cells. The molecular weight of FABP4 is approximately 14-15 kDa. It exhibits high expression levels in adipose tissue and is inducible in macrophages under certain conditions.
Biological function summary

This protein participates in the regulation of lipid homeostasis and energy metabolism. FABP4 is part of the lipid-binding protein family that helps maintain fatty acid equilibrium within cells.
