Activity summary

Factor D also known as complement factor D or CFD protein is a serine protease involved in the alternative pathway of the complement system. It plays a mechanical role by cleaving factor B when it is bound to C3b forming the active C3 convertase C3bBb. This enzyme is relatively small with a mass of approximately 28.5 kDa. It is mainly expressed in adipose tissue but it can also be found in the blood plasma.

Biological function summary

Factor D aids the immune system's ability to destroy pathogens. As part of the alternative complement pathway it acts independently and does not associate with larger protein complexes. Its activation of the alternative pathway leads to direct lysis of pathogens therefore contributing to innate immunity.

Pathways

Factor D participates specifically in the complement activation pathway particularly in the innate immune response. It works closely with other proteins in the complement system such as factor B and C3. This pathway enhances the opsonization and removal of microbes and damaged cells acting in parallel with the classical pathway of the complement system.

Associated diseases and disorders

Abnormal function or regulation of factor D can relate to chronic inflammation and age-related macular degeneration. Disorders involving factor D may involve excess complement activation leading to tissue damage. The role of factor D in these diseases often connects it to other key complement components such as C3 and factor B which can also become problematic when dysregulated.
