Immunocytochemistry/ Immunofluorescence - Anti-FAK antibody [RP23040020] (AB307996)
For immunofluorescence analysis, 70% confluent log phase A549 cells were fixed with 4% paraformaldehyde for 15 minutes; permeabilized with 0.25% Triton X-100 for 10 minutes followed by blocking with 5% BSA for 1 hour at room temperature. To detect FAK, cells were labelled using ab307996 at 2-4 µg in 1% BSA and incubated for 3 hours at room temperature and then labeled with Alexa Fluor® 488 Goat anti-Rabbit IgG Secondary Antibody at a dilution of 1/400 for 30 minutes at room temperature (Panel a : green). Nuclei (Panel b : blue) were stained with DAPI. F-actin (Panel c : red) was stained with Alexa Fluor® 594 Phalloidin. Panel d is a merged image showing cytoplasmic and punctate staining on the membrane. The images were captured at 20X magnification.

Western blot - Anti-FAK antibody [RP23040020] (AB307996)
Western blot analysis of FAK was performed by loading Jurkat (lane1) and A549 (lane2) cell lysates using 4-12 % Bis-Tris gel, electrophoresis system, and pre-stained protein standard. Proteins were transferred to a PVDF membrane and blocked with 5 % skim milk for 1 hour at room temperature. FAK was detected at ~125 kDa using ab307996 at 1-3 µg/mL in 2.5 % skim milk at 4°C overnight on a rocking platform. Goat anti-Rabbit IgG-HRP Secondary Antibody at 1 : 5000 dilution was used and chemiluminescent detection was performed (ECL).

Biological function summary
Focal Adhesion Kinase participates in the formation of focal adhesions which are complexes that connect the cytoskeleton to the extracellular matrix. The FAK protein functions as an important signaling node in these structures allowing for the assembly of multiprotein signal transduction complexes. FAK also controls cellular processes such as spreading motility and survival. The interaction with proteins such as Src kinases paxillin and talin facilitates its biological roles in cell signaling.
Pathways
Focal Adhesion Kinase engages in the regulation of the MAPK/ERK signaling pathway and the PI3K/AKT pathway. These pathways are instrumental for cell proliferation survival and migration. In these pathways FAK interacts with proteins such as PI3K Grb2 and Sos linking integrin-mediated signals with downstream effects that influence cell behavior and survival.
