The protein FAM134B also known as JK-1 is an autophagy regulator with a molecular mass of approximately 50 kDa. FAM134B plays a critical role in the degradation of endoplasmic reticulum (ER) membranes. This protein expresses robustly in the neural tissues gastrointestinal tract and reproductive organs. FAM134B functions by binding to membranes and facilitating the removal of damaged ER components which maintains cellular homeostasis.
FAM134B serves as an important player in maintaining cell health by its involvement in ER-phagy a selective autophagy process. It forms part of the ER-phagy complex and works alongside LC3 proteins to ensure efficient turnover of the ER membrane. Its activity helps prevent ER stress and safeguard cell survival under stress conditions. Failure in this pathway can lead to accumulation of misfolded proteins and impaired cell function.
FAM134B interacts in the process of ER-associated degradation (ERAD) and autophagy-related pathways. It participates importantly in the calcium signaling pathway where FAM134B affects the homeostasis of calcium levels in the cytoplasm and ER. This interaction involves key signaling proteins such as ATG8 and ER-phagy receptors showing its essential role in cellular adaptation to stress.
FAM134B has significant associations with sensory neuropathy and colorectal cancer. In sensory neuropathy mutations or dysfunctions in FAM134B lead to the loss of pain perception due to improper ER turnover in neurons. It also links to colorectal cancer often showing altered expression that affects tumor progression. FAM134B interacts with proteins like p53 in cancer pathways which influences cell cycle regulation and apoptosis in cancerous cells.

FAM134B was immunoprecipitated from 0.35 mg HeLa (human cervical adenocarcinoma epithelial cell) whole cell lysate with ab316161 at 1/30 dilution (2ug in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab316161 at 1/1000 dilution. VeriBlot for IP secondary antibody(HRP)(VeriBlot for IP Detection Reagent (HRP) ab131366) was used at 1/5000 dilution.
