Function

Endoplasmic reticulum (ER)-anchored autophagy regulator which exists in an inactive state under basal conditions but is activated following cellular stress (PubMed:34338405). When activated, induces ER fragmentation and mediates ER delivery into lysosomes through sequestration into autophagosomes via interaction with ATG8 family proteins (PubMed:34338405). Promotes ER membrane curvature and ER tubulation required for subsequent ER fragmentation and engulfment into autophagosomes (PubMed:33826365). Required for collagen quality control in a LIR motif-dependent manner (By similarity). Mediates NRF1-enhanced neurite outgrowth (PubMed:26040720).

Activity summary

FAM134C also known as family with sequence similarity 134 member C is a protein involved in autophagy processes. It has a molecular mass of approximately 54 kDa. This protein localizes mainly within the endoplasmic reticulum (ER) membrane and it is expressed in several tissues including the brain and liver. FAM134C plays an essential role in regulating the ER shape and maintaining cellular homeostasis.

Biological function summary

The protein participates in the process of selective autophagy particularly ER-phagy which is the degradation of ER fragments. This function indicates that it is part of a broader complex that regulates the destruction of damaged or excessive organelles which is important for neuron and liver cell function. FAM134C interacts with LC3 proteins during these processes acting as a receptor that identifies ER regions targeted for degradation.
