This data was developed using ab307591, the same antibody clone in a different buffer formulation. Flow cytometric analysis of human peripheral blood mononuclear cell (PBMC) cells labeling Fas with ab307591 at 1/50 dilution (1µg) (Right panel) compared with a Rabbit monoclonal IgG (ab172730) isotype control (Left panel). Goat Anti-Rabbit IgG (Alexa Fluor® 488, ab150081) at 1/2000 dilution was used as the secondary antibody. Gated on viable cells. Cells were stained with rabbit IgG or ab307591, then stained with anti-CD45RO conjugated to BV421.

Flow cytometric analysis of K562 (human chronic myelogenous leukemia lymphoblast) (Left panel) / Jurkat (human T cell leukemia T lymphocyte) (Right panel) cells labeling Fas with ab307591 at 1/50 dilution (1µg) (Red) compared with a Rabbit monoclonal IgG (ab172730) (Black) isotype control and an unlabeled control (cells without incubation with primary antibody and secondary antibody) (Blue). Goat Anti-Rabbit IgG (Alexa Fluor® 488, ab150081) at 1/2000 dilution was used as the secondary antibody. Gated on viable cells. Low expression : K-562 (PMID : 9168303).

Target data

Receptor for TNFSF6/FASLG. The adapter molecule FADD recruits caspase CASP8 to the activated receptor. The resulting death-inducing signaling complex (DISC) performs CASP8 proteolytic activation which initiates the subsequent cascade of caspases (aspartate-specific cysteine proteases) mediating apoptosis. FAS-mediated apoptosis may have a role in the induction of peripheral tolerance, in the antigen-stimulated suicide of mature T-cells, or both. The secreted isoforms 2 to 6 block apoptosis (in vitro).
