FBN3 also known as Fibrillin-3 is a glycoprotein with a mass of approximately 350 kDa. It belongs to the fibrillin family of proteins which are essential components of the extracellular matrix. FBN3 is predominantly expressed in the fetal and adult brain as well as the adrenal gland. These expression patterns suggest its potential role in neural development and function although the full spectrum of its expression remains to be completely defined.
Biological function summary

FBN3 plays a major role in the structure and function of connective tissues. It often combines with other fibrillin proteins to form microfibrils. Microfibrils contribute to the structural integrity and elasticity of tissues. The ability of FBN3 to interact with other components of the extracellular matrix including elastin positions it as an important element in maintaining the physical properties of tissues especially those requiring resilience such as skin and heart tissues.

Pathways

FBN3 participates in the TGF-beta signaling pathway which regulates cell growth proliferation and differentiation. The protein interacts with other members of the fibrillin family especially FBN1 and FBN2 within these pathways. Its engagement in such pathways highlights its role in regulating extracellular matrix formation. Also FBN3 contributes to maintaining tissue homeostasis highlighting its significant part in developmental processes.

Alterations in FBN3 expression or function link to Marfan syndrome a genetic disorder affecting connective tissue integrity and idiopathic scoliosis a condition characterized by abnormal spine curvature. Similar to its family members FBN3 interacts with FBN1 affecting connective tissue properties in these diseases. Understanding FBN3's role opens potential therapeutic avenues for these and possibly other connective tissue disorders.
