May act as a link between RND2 signaling and regulation of the actin cytoskeleton (By similarity). Required to coordinate membrane tubulation with reorganization of the actin cytoskeleton during the late stage of clathrin-mediated endocytosis. Binds to lipids such as phosphatidylinositol 4,5-bisphosphate and phosphatidylserine and promotes membrane invagination and the formation of tubules. Also enhances actin polymerization via the recruitment of WASL/N-WASP, which in turn activates the Arp2/3 complex. Actin polymerization may promote the fission of membrane tubules to form endocytic vesicles. May be required for the lysosomal retention of FASLG/FASL.

FBP17 formally known as Formin-binding protein 17 is a protein with a molecular mass of approximately 59 kDa. It features an F-BAR domain that facilitates membrane bending and cytoskeletal organization. Expressed mainly in neural tissues FBP17 affects the dynamics of the cell membrane and is involved in the formation of tubular structures critical for various cellular processes. Researchers classify it as an important player in endocytosis where its role is mechanical and structural within the cell.
In cellular environments where signal transduction is occurring FBP17 engages in the regulation of actin polymerization. It participates in complex formations with other proteins like Bin2 and is important for the recruitment of actin nucleators. These interactions help in directing changes to cellular structure during activities such as migration and division. FBP17 can also influence the intracellular trafficking of signals further impacting how cells react to environmental stimuli.
Where cell interactions and signaling occur FBP17 integrates into the pathways of actin cytoskeleton reorganization and endocytosis. It works alongside proteins like Cdc42 to modulate these pathways contributing significantly to cellular morphology adjustments and the uptake of external materials. In these processes changes in actin dynamics facilitated by FBP17 and its pathway partners provide the cellular capacity for movement and the intake of necessary substances.
Wherein cellular malfunctions manifest FBP17 has been linked to neurodegenerative diseases including Charcot-Marie-Tooth disease. In this context disruptions in FBP17 functions can alter cellular homeostasis leading to neurodegeneration. Additionally FBP17 associates with amphiphysin impacting stress response pathways and signaling which are often implicated in cancer. Understanding FBP17's role may offer insights into these diseases and guide therapeutic strategies.
